Ioannes 44 was bishop of Amaseia in Helenopontus; in 681 he attended the Third Council of Constantinople (the Sixth Ecumenical Council) and was present at two of the last three sessions (sessions sixteen and eighteen, on 9 August and 16 September): Riedinger II 2. 686-823 (= Mansi XI 613-672). In the subscriptions to the statement of the faith, which condemned monotheletism, and to the Council, he is styled Ἰωάννης ἐλάχιστος ἐπίσκοπος (Riedinger II 2. 823, line 23 (= Mansi XI 672) reads: ἐλέει Θεοῦ ἐπίσκοπος) τῆς Ἀμασέων μητροπόλεως τῆς Ἑλενοποντίων ἐπαρχίας: Riedinger II 2. 782, line 3, 823, line 23, cf. II 2. 892, line 17 (= Mansi XI 641, 672, cf. XI 689). The old Latin version printed in Riedinger II 2. 783, line 3, calls his see "Amasiae metropoleos Helenoponti". In the lists of those attending each session he is styled Ἰωάννου ἐπισκόπου Ἀμασείας: Riedinger II 2. 686, line 1, 756, line 7 (= Mansi XI 613, 625) (at Riedinger II 2. 756 (= Mansi XI 625) the entries for the bishops of Amaseia and Side have been run together, confirmed by the Latin text at Riedinger II 2. 757 (= Mansi XI 626); see Ioannes 37). He was among the signatories to the copy of the definition of the faith sent to the bishop of Rome, Agatho 1, signing on behalf of himself and the bishops under him: Riedinger II 2. 892, line 17 (= Mansi XI 689) (ὑπὲρ ἐμαυτοῦ καὶ τῆς ὑπ'ἐμὲ συνόδου). He was still bishop of Amaseia in 692, when he attended the Quinisext Council and signed the canons: Ohme, Bischofsliste, p. 147, no. 23 (= Mansi XI 989) (Ἰωάννης ἀνάξιος ἐπίσκοπος τῆς Ἀμασέων μητροπόλεως τῆς Ἑλενοποντίων ἐπαρχίας).
