Antonios 6 was a monk; in 787 he attended the Second Council of Nikaia (the Seventh Ecumenical Council) as representative of the see of Smyrna; he is attested at the fourth and seventh sessions, on 1 October and 13 October 787, and he was perhaps present at the last five sessions at least, from 1 October to 23 November 787: Mansi XIII 137-486. The see was represented at the first two sessions by Gregorios 23. In the list of those present at the seventh session he is styled Ἀντωνίου μοναχοῦ καὶ τοποτηρητοῦ Σμύρνης: Mansi XIII 365. Elsewhere he is styled Ἀντώνιος ἀνάξιος μοναχὸς καὶ ἐκ προσώπου τοῦ θρόνου Σμύρνης, Mansi XIII 137; and Ἀντώνιος ἁμαρτωλὸς μοναχὸς καὶ τοποτηρητὴς τοῦ θρόνου Σμύρνης, Mansi XIII 384. At the fourth session he subscribed the statements read out from the Fathers in support of the veneration of icons: Mansi XIII 137. At the seventh session he subscribed the statement of the faith adopted by the Council: Mansi XIII 384.
